PLEASE NOTE - As far as I know, this only occurs in humans. But I thought it would be cool to have a lethal mutation doesn't kill a lion before it grows out of it's cub stage.
Infantile neuroaxonal dystrophy (INAD) is a rare inherited neurological disorder. It affects axons, the part of a nerve cell that carries messages from the brain to other parts of the body, and causes progressive loss of vision, muscular control, and mental skills. .... Distinctive facial deformities may be present at birth, including a prominent forehead, crossed eyes, an unusually small nose or jaw, and large, low-set ears. INAD is a progressive disease. Once symptoms begin, they will worsen over time. .... Rapid, wobbly eye movements and squints may be the first symptoms, followed by floppiness in the body and legs (more than in the arms). Death usually occurs between the ages of 5 to 10 years.

How INAD MIGHT Work In-Game
This mutation, or rather, neurological disorder, would be present from 0 Months all the way up to 4 Years old. Males cannot become kings, they can however become submales, but they cannot patrol. Females on the other hand will be able to breed up until 3 Years. As at 4 years old, I have them planned on dying.

6 Months
*Cub* seemed be doing well before. But your lioness notices that *Cub* is learning things slower then the others.

1 Year
Your lioness is happy to see *Cub* made it to it's adolescent years! Even though *Cub* is still learning slower then the rest of her/his siblings. *Cub* seems to be doing okay.

1 Year, 7 Months
*Cub* seems to be having difficulties with her/his vision as well seeming to loose some of her/his skills. She/He seems to be squinting more often then usual. She/He even runs into things if they aren't being careful enough. Some of the other lions/lionesses laugh at him/her, finding it funny. But now you've grown concerned about *Cub's* health.
[Possible slower gained stats? Possible no stats gained?]

2 Years
*Cub* has made it to adult! However, your concern grows more for her/him because they are having a difficult time standing and holding their head up. They can not hunt/patrol.
[Unable To Patrol/Hunt?]

3 Years
All *Cub* does is lay about the den. She/He seems to be mostly blind and she/he can only stand for so long before she/he needs to rest for the remainder of the day. And even then, they typically tend to stay to one corner of the den.

3 Years, 5 Months
*Cub* seems to have grown worse in the last few days. They doesn't seem to eat or talk much. You're worried he/she won't make it for very long.

4 Years
*Cub* died last night due to having a neurological disorder.

i'd add that the females cant hunt and the males cant patrol, being blind and having trouble moving could reasonably do that

yeah, it makes more sense to me that they wouldnt be able to hunt/patrol at all, especially since the way you described it makes it seem like the onset for more debilitating symptoms manifest early on

Makes sense. That would mean she/he would only make it until 3 or 4 years old. Which, I suppose is reasonable considering what happens. And would give a decent amount of time for breeding.

Good thinking though. I'll edit this tonight then with those changes.

I support! I like the idea of this new "mutation"! Here are my ideas:
6 Months: Slower gained stats
1 Year: Slower gained stats
1 Year 7 Months: Maybe gain some stats slowly and loose some randomly?
2 Years: Unable to hunt and patrol.
