For a first generation tigon it's basically just dumb luck - they're just so rare that it's very unlikely to ever breed one. You can increase your chances a bit by using mutation items like CRBs or GMO cows, but even then having a hybrid pop up is extremely extremely rare (hence why first gens are so few and far between, and so valuable).

The vast majority of tigons in the game are not bred that way, but rather are second+ generation - meaning they are descended from existing tigon mothers. That increases your odds exponentially, because female tigons have a 5% chance per cub of naturally passing it down. Or that's doubled to a 10% chance per cub if the mother is a first generation tigon.

So generally by far the most effective way to get one is to buy a tigon and then keep breeding from her. Or if you don't have enough to afford that upfront, then to pay for reverse heats from someone else's tigon and hope to breed one for less expenditure that way. Obviously using a buffalo scrotum is advised, since that improves your chances a lot because it maximises the number of cubs produced :)
